Descrizione Molecular Identification (16S rRNA Gene) of Amylase-Producing Bacteria

Amylase is an enzyme that catalyzes the hydrolysis of the ¿-1,4-glycosidic bond on starch to glucose. Amylases derived from thermophile bacteria are widely used in industry because they are stable against pH and high temperatures, such as bacteria originating from hot springs. Semurup hot spring has a temperature of 80°C, pH 8.4, contains various organic, inorganic and mineral compounds. This leads to high bacteria was diversity at Semurup hot spring. The aim of the study were to obtain isolates of amylase-producing bacteria with the highest activity, stabilize temperature, and determine bacteria strains of thermophile bacteria originating from Semurup hot springs. Methods of research have been done, namely isolation and screening of amylase-producing bacteria, bacteria identification. The result of isolation and screening thermophile bacteria obtained 24 isolates of amylase-producing bacteria. Bacteria isolate SMR1.1 is one of the 24 isolates of amylase-producing bacteria that have the highest activity, stable against the temperature. The result of identification showed that isolate of SMR1.1 bacteria belong to new strain bacteria, that is Bacillus licheniformis strain Megasis
