Solid tumours differ from normal tissue in a numberof important ways. Rapid growth and metabolism oftumours often leads to altered physiology includingchanges in vasculature and perfusion which can inturn induce fluctuations in the supply of nutrientssuch as oxygen. Reduced tissue oxygenation, known ashypoxia, has been linked to radio- andchemotherapeutic resistance and is often associatedwith a poor prognosis for disease-free survival incancer patients. Identification of tumour hypoxia by usingnon-invasive imaging techniques such as positronemission tomography (PET) is vital for clinicians toprovide effective treatment. However, physiologicalimaging modalities such as PET require the design andadministration of radioactive imaging agents whichspecifically target tumours. This book provides a background to the basic chemicalconcepts and methods used in the rational design oftumour-selective radiopharmaceuticals with particularemphasis on the mechanisms by whichcopper-based imaging agents target tumour hypoxia.
